(AP) ? The NHL is set to get back to the bargaining table with the locked-out players' association after a new contract offer from the league broke the ice between the sides.

Those talks are scheduled to take place Sunday in New York, nearly one month after negotiations broke down and pushed the sides further apart.

Without fanfare, the NHL said Friday that it made a new proposal to the union on Thursday in an effort to end the lockout and save the delayed hockey season.

The players' association was still reviewing the document, which was several hundred pages in length, on Friday night and planned to speak to the NHL by telephone on Saturday, and then get together with the league on Sunday.

The sides haven't met since a second round of talks with a federal mediator ended on Dec. 13. They haven't had formal negotiations alone since the first week of the month.

Time is running out to come to an agreement and then play a season of at least 48 games ? a number the NHL set as a minimum. The league would like to reach a deal by Jan. 11, start training camp the following day, and open the season by Jan. 19.

"We delivered to the union a new, comprehensive proposal for a successor CBA," NHL deputy commissioner Bill Daly said in a statement Friday. "We are not prepared to discuss the details of our proposal at this time. We are hopeful that once the union's staff and negotiating committee have had an opportunity to thoroughly review and consider our new proposal, they will share it with the players. We want to be back on the ice as soon as possible."

The players' association's executive board and negotiating committee went over the proposal during an internal conference call on Friday, the 104th day of the lockout.

A person familiar with key points of the offer told The Associated Press that the league proposed raising the limit of individual free-agent contracts to six years from five ? seven years if a team re-signs its own player; raising the salary variance from one year to another to 10 percent, up from 5 percent; and one compliance buyout for the 2013-14 season that wouldn't count toward a team's salary cap but would be included in the overall players' share of income.

The person spoke on condition of anonymity because details of the new offer were not being discussed publicly.

The NHL maintained the deferred payment amount of $300 million it offered in its previous proposal, an increase from an earlier offer of $211 million. The initial $300 million offer was pulled off the table after negotiations broke off earlier this month.

The latest proposal is for 10 years, running through the 2021-22 season, with both sides having the right to opt out after eight years.

The lockout has reached a critical stage, threatening to shut down a season for the second time in eight years. All games through Jan. 14, plus the Winter Classic and the All-Star game already have been called off. The next round of cuts could claim the entire schedule.

The NHL is the only North American professional sports league to cancel a season because of a labor dispute, losing the 2004-05 campaign to a lockout. A 48-game season was played in 1995 after a lockout stretched into January.

It is still possible this dispute could eventually be settled in the courts if the sides can't reach a deal on their own.

The NHL filed a class-action suit this month in U.S. District Court in New York in an effort to show its lockout is legal. In a separate move, the league filed an unfair labor practice charge with the National Labor Relations Board, contending bad-faith bargaining by the union.

Those moves were made because the players' association took steps toward potentially filing a "disclaimer of interest," which would dissolve the union and make it a trade association. That would allow players to file antitrust lawsuits against the NHL.

Union members voted overwhelmingly to give their board the power to file the disclaimer by Jan. 2. If that deadline passes, another authorization vote could be held to approve a later filing.

Negotiations between the NHL and the union have been at a standstill since talks ended Dec. 6. One week later, the sides convened again with federal mediators in New Jersey, but still couldn't make progress.

The sides have been unable to reach agreement on the length of the new deal, the length of individual player contracts, and the variance in salary from year to year. The NHL is looking for an even split of revenues with players.

The NHL pulled all previous offers off the table after the union didn't agree to terms on its last proposal without negotiation.

What worked for me is just turning off Google Now. Yeah you lose some functionality, but it doubles your battery life. Google really screwed up with this one. I really don't know who to blame, Google, Samsung, or Verizon. I dont get these wakelocks on my Nexus with the same app, so it might be something to do with what Samsung did with TW. No wakelocks on 4.2 either.

Genetic sequencing breakthrough to aid treatment for congenital hyperinsulinism

Dec. 27, 2012 ? Congenital hyperinsulinism is a genetic condition where a baby's pancreas secretes too much insulin. It affects approximately one in 50,000 live births and in severe cases requires the surgical removal of all or part of the pancreas.

Researchers at the University of Exeter Medical School are the first in the world to utilise new genetic sequencing technology to sequence the entirety of a gene in order to identify mutations that cause hyperinsulinism. Previously, existing technology limited such sequencing to only part of the coding regions of the gene which meant that some mutations were missed.

Using new Illumina genetic sequencing technology, the research team led by Professor Sian Ellard has discovered novel mutations that cause hyperinsulinism. Their findings are published December 27 2012, on-line by The American Journal of Human Genetics.

The outcome will be that some infants born with hyperinsulinism will require fewer investigations, because the new technology means that for many only one genetic test will be required to determine the extent of the condition in each child. It also means that clinicians will have more information at their fingertips to inform them about how much of the pancreas needs to be removed.

Approximately 50 per cent of patients with congenital hyperinsulinism require surgery, and of those half require the entire pancreas to be removed. Removal of the entire pancreas increases the risk of diabetes later in life, but if left undiagnosed and untreated hyperinsulinism can result in irreparable brain damage. Symptoms range from shakiness and tiredness to seizure and coma.

Dr. Sarah Flanagan, Research Fellow in Molecular Genetics at the University of Exeter Medical School said: "The potential provided by this new technology is important and exciting, because it allows us to investigate genetic coding in its entirety. This means that investigators can identify mutations that sit at the heart of any number of conditions where before they might have been missed. This in turn results in better information for clinicians upon which they can base effective treatments and interventions for their patients."

Strange behavior: New study exposes living cells to synthetic protein

Dec. 27, 2012 ? One approach to understanding components in living organisms is to attempt to create them artificially, using principles of chemistry, engineering and genetics. A suite of powerful techniques -- collectively referred to as synthetic biology -- have been used to produce self-replicating molecules, artificial pathways in living systems and organisms bearing synthetic genomes.

In a new twist, John Chaput, a researcher at Arizona State University's Biodesign Institute and colleagues at the Department of Pharmacology, Midwestern University, Glendale, AZ have fabricated an artificial protein in the laboratory and examined the surprising ways living cells respond to it.

"If you take a protein that was created in a test tube and put it inside a cell, does it still function," Chaput asks. "Does the cell recognize it? Does the cell just chew it up and spit it out?" This unexplored area represents a new domain for synthetic biology and may ultimately lead to the development of novel therapeutic agents.

The research results, reported in the advanced online edition of the journal ACS Chemical Biology, describe a peculiar set of adaptations exhibited by Escherichia coli bacterial cells exposed to a synthetic protein, dubbed DX. Inside the cell, DX proteins bind with molecules of ATP, the energy source required by all biological entities.

"ATP is the energy currency of life," Chaput says. The phosphodiester bonds of ATP contain the energy necessary to drive reactions in living systems, giving up their stored energy when these bonds are chemically cleaved. The depletion of available intracellular ATP by DX binding disrupts normal metabolic activity in the cells, preventing them from dividing, (though they continue to grow).

After exposure to DX, the normally spherical E. coli bacteria develop into elongated filaments. Within the filamentous bacteria, dense intracellular lipid structures act to partition the cell at regular intervals along its length. These unusual structures, which the authors call endoliposomes, are an unprecedented phenomenon in such cells.

"Somewhere along the line of this filamentation, other processes begin to happen that we haven't fully understood at the genetic level, but we can see the results phenotypically," Chaput says. "These dense lipid structures are forming at very regular regions along the filamented cell and it looks like it could be a defense mechanism, allowing the cell to compartmentalize itself." This peculiar adaptation has never been observed in bacterial cells and appears unique for a single-celled organism.

Producing a synthetic protein like DX, which can mimic the elaborate folding characteristics of naturally occurring proteins and bind with a key metabolite like ATP is no easy task. As Chaput explains, a clever strategy known as mRNA display was used to produce, fine-tune and amplify synthetic proteins capable of binding ATP with high affinity and specificity, much as a naturally occurring ATP-binding protein would.

First, large libraries of random sequence peptides are formed from the four nucleic acids making up DNA, with each strand measuring around 80 nucleotides in length. These sequences are then transcribed into RNA with the help of an enzyme -- RNA polymerase. If a natural ribosome is then introduced, it attaches to the strand and reads the random sequence RNA as though it was a naturally-occurring RNA, generating a synthetic protein as it migrates along the strand. In this way, synthetic proteins based on random RNA sequences can be generated.

Exposing the batch of synthetic proteins to the target molecule and extracting those that bind can then select for ATP-binding proteins. But as Chaput explains, there's a problem: "The big question is how do you recover that genetic information? You can't reverse transcribe a protein back into DNA. You can't PCR amplify a protein. So we have to do all these molecular biology tricks."

The main trick involves an earlier step in the process. A molecular linker is chemically attached to the RNA templates, such that each RNA strand forms a bond with its newly translated protein. The mRNA-protein hybrids are exposed to selection targets (like ATP) over consecutive rounds of increasing stringency. After each round of selection, those library members that remain bound to the target are reverse-transcribed into cDNA (using their conveniently attached RNA messages), and then PCR amplified.

In the current study, E. coli cells exposed to DX transitioned into a filamentous form, which can occur naturally when such cells are subject to conditions of stress. The cells display low metabolic activity and limited cell division, presumably owing to their ATP-starved condition.

The study also examined the ability of E. coli to recover following DX exposure. The cells were found to enter a quiescent state known as viable but non-culturable (VBNC), meaning that they survived ATP sequestration and returned to their non-filamentous state after 48 hours, but lost their reproductive capacity. Further, this condition was difficult to reverse and seems to involve a fundamental reprogramming of the cell.

In an additional response to DX, the filamentous cells form previously undocumented structures, which the authors refer to as endoliposomes. These dense lipid concentrations, spanning the full width of the filamented E. coli, segment the cells into distinct compartments, giving the cells a stringbean-like appearance under the microscope.

The authors speculate that this adaptation may be an effort to maintain homeostasis in regions of the filamentous cell, which have essentially been walled off from the intrusion of ATP-depleting DX. They liken endoliposomes to the series of water-tight compartments found in submarines which are used to isolate damaged sections of the ship and speculate that DX-exposed cells are partitioning their genetic information into regions where it can be safely quarantined. Such self-compartmentalization is known to occur in some eukaryotic cells, but has not been previously observed in prokaryotes like E. coli.

The research indicates that there is still a great deal to learn about bacterial behavior and the repertoire of responses available when such cells encounter novel situations, such as an unfamiliar, synthetic protein. The study also notes that many infectious agents rely on a dormant state, (similar to the VBNC condition observed in the DX-exposed E. coli), to elude detection by antibiotics. A better understanding of the mechanisms driving this behavior could provide a new approach to targeting such pathogens.

The relative safety of E. coli as a model organism for study may provide a fruitful tool for more in-depth investigation of VBNC states in pathogenic organisms. Further, given ATP's central importance for living organisms, its suppression may provide another avenue for combating disease. One example would be an engineered bacteriophage capable of delivering DX genes to pathogenic organisms.

LAGOS, Nigeria (AP) ? A massive explosion ripped through a warehouse full of fireworks in Nigeria's largest city on Wednesday, sparking a fire that threatened surrounding city blocks and sending a plume of thick smoke high into the sky. At least one person died and 15 others were wounded, emergency officials said.

The blast occurred around 9 a.m. in the Jankara area of Lagos Island, a neighborhood of narrow streets and tall cement buildings holding shops and housing people sometimes a dozen to a room in the megacity of Lagos. The force of the explosion echoed miles away and shook windows.

An Associated Press journalist saw members of the Nigerian Red Cross treating people with minor cuts and bruises a few blocks from the site. Later, rescuers pulled out a badly charred corpse from the still-smoldering structure.

Many people were injured when they stampeded through the area's narrow alleyways, said Yushau Shuaib, a spokesman for Nigeria's National Emergency Management Agency.

"The explosion was not suspected to be a deliberate attack to cause confusion as security agencies have commenced their investigations," Shuaib said in a statement later Wednesday.

Casualties, however, could rise as rescuers continue to clear debris from the fire. Smoke still rose over the neighborhood as the sun started to set Wednesday over Lagos.

Rubble-strewn streets surrounding the burning buildings were covered in the spent shells of fireworks. Several police officers and security officials found large, mortar-like fireworks in the building, the kind wired to explode as part of a large arrangement put on by professionals.

Nigerians commonly shoot off fireworks and firecrackers to mark Christmas and the New Year, with hawkers roaming in traffic with fistfuls of the small explosives to sell to passing drivers.

Local government officials have tried to ban fireworks this year, in part due to safety and in part due to the fear they can cause in a nation where a radical Islamist sect is carrying out shootings and car bombings. Nevertheless fireworks have exploded across the country in recent days to mark the holiday season.

Immediately after the blast Wednesday, thousands quickly filled the streets surrounding the burning buildings. Others ran away as fireworks continued to explode inside. A half-dozen firefighters arrived at the scene with two trucks and locals also ran fire hoses from the trucks to nearby buildings to try to beat back the flames.

The trucks quickly ran out of water. One man even scooped up water from a puddle with a bowl in an attempt to fight the blaze. One firefighter briefly fought with a young man who took his firefighting helmet off his head and ran away into the swarming crowd. The firefighter shrugged and went back to his truck as the building continued to burn.

Breast cancer may be on the increase. ?But much less than one might think, given all the publicity. ?Most of the scary increase in breast cancer is actually due to early stage detection through mamograms of conditions that may never progress to invasive cancer. ?Plus, there is a ton and a half of media hype generated by huge charitable and commercial?industries?that feed on the cancer epidemic - be it real or not - making the problem seem much worse than it really is.

Why does your doctor treat all these "positives" so aggressively?

Cancer is a terrible disease that I would not wish upon my worst enemy. ?One of my aunts passed away a few years ago, some 15 years after discovering she had breast cancer (She opted for no treatment, a decision I did not agree with at the time). ?She enjoyed over a decade of good health before finally?succumbing?to its ravages.

The problem with breast cancer is it may be next to impossible to know which of these many false positives will go on to be invasive cancer.
If there is an aggressive cancer present, then the sooner it is treated - aggressively - the better the chances of survival. ?
If your doctor decides to take a "wait and see" approach and it consequently develops into an aggressive cancer, he/she has got a serious problem - a very unhappy patient who may complain to the authorities and even sue for negligence. ?Better to play it safe, professionally, and order the full works for the patient. ?No doctor ever got sued for over-treating a breast lump (as far as I know anyway). ?And no doctor ever wants to lose a patient through "under treatment".

If you have a breast lump, what should you do?

Here is what I would do:

  • Prevention is best: Get a thermogram. ?A thermogram, unlike a mamogram, is totally non-invasive and is valid for all women, young and old. A number of my clients have had this done and the results are most useful - and reassuring. ?Go here for more information.
  • Do not fiddle with it - that means, leave it alone! ?Do not poke and prod and do not crush it with a mamogram or allow a biopsy at this stage. ?You do not want to break up any tumour tissue, causing cells to migrate to other parts of the body. ?As an aside: a mamogram is ionising radiation which increases your lifetime risk of getting cancer, so I would be inclined to use less invasive and less potentially harmful assessment tools in the early stages of investigation.
  • Get a thermogram asap, or a new one if you have had one in the past, for comparison (repeat thermograms are extremely valuable because they will show any changes that may have happened). ?This is best done directly via a thermogram screening service. A thermogram is totally non-invasive. ?The thermogram will pick up the heat signature, in remarkable detail, of vascularisation associated with a growing tumour.

To reduce mortality, screening must detect life-threatening disease at an earlier, more curable stage. Effective cancer-screening programs therefore both increase the incidence of cancer detected at an early stage and decrease the incidence of cancer presenting at a late stage.

METHODS

We used Surveillance, Epidemiology, and End Results data to examine trends from 1976 through 2008 in the incidence of early-stage breast cancer (ductal carcinoma in situ and localized disease) and late-stage breast cancer (regional and distant disease) among women 40 years of age or older.

RESULTS

The introduction of screening mammography in the United States has been associated with a doubling in the number of cases of early-stage breast cancer that are detected each year, from 112 to 234 cases per 100,000 women ? an absolute increase of 122 cases per 100,000 women. Concomitantly, the rate at which women present with late-stage cancer has decreased by 8%, from 102 to 94 cases per 100,000 women ? an absolute decrease of 8 cases per 100,000 women. With the assumption of a constant underlying disease burden, only 8 of the 122 additional early-stage cancers diagnosed were expected to progress to advanced disease. After excluding the transient excess incidence associated with hormone-replacement therapy and adjusting for trends in the incidence of breast cancer among women younger than 40 years of age, we estimated that breast cancer was overdiagnosed (i.e., tumors were detected on screening that would never have led to clinical symptoms) in 1.3 million U.S. women in the past 30 years. We estimated that in 2008, breast cancer was overdiagnosed in more than 70,000 women; this accounted for 31% of all breast cancers diagnosed.

CONCLUSIONS

Despite substantial increases in the number of cases of early-stage breast cancer detected, screening mammography has only marginally reduced the rate at which women present with advanced cancer. Although it is not certain which women have been affected, the imbalance suggests that there is substantial overdiagnosis, accounting for nearly a third of all newly diagnosed breast cancers, and that screening is having, at best, only a small effect on the rate of death from breast cancer.

Sacha Baron Cohen 'failed' first 'Les Mis' audition

Singing is a perfect way to ring in Christmas Day, but who knew it would be coming from TODAY's Matt Lauer and "Les Miserables" star Sacha Baron Cohen?

Baron Cohen spoke with Lauer on Tuesday, but not before the actor began crooning, "I am contractually obligated to do this whole interview in song." Baron Cohen plays?corrupt innkeeper Monsieur Thenardier in the film, and like all of the actors, sings all of his lines in the movie.

He noted (after returning to his usual spoken voice) that "Les Miserables" is "a staple in London," and when he was 20, he gave a stab at being in the chorus. "I passed the singing bit," he noted, but fell down when it came to the dancing part. Not that he didn't give it a good try: "There's like 200 people standing outside in the rain (at the audition), and then it came to the dancing bit ... and you had to learn a routine and it was clear I had absolutely no idea how to do it," said Baron Cohen. "So when it came time for action ... I decided to freestyle."?

He did not get the job. "I failed," he added.

Not so in "Les Miserables," where there's a lot of singing (and minimal dancing). By the end of the interview, Baron Cohen was back to singing -- and so was Lauer. Check out the video for the rest!

"Les Miserables" opened in theaters on Dec. 25.

Dental assistant fired for being 'irresistible' is 'devastated'

After working as a dental assistant for ten years, Melissa Nelson was fired for being too "irresistible" and a "threat" to her employer's marriage.

"I think it is completely wrong," Nelson said. "I think it is sending a message that men can do whatever they want in the work force."

On Friday, the all-male Iowa State Supreme Court ruled that James Knight, Nelson's boss, was within his legal rights when he fired her, affirming the decision of a lower court.

"We do think the Iowa Supreme Court got it completely right," said Stuart Cochrane, an attorney for James Knight. "Our position has always been Mrs. Nelson was never terminated because of her gender, she was terminated because of concerns her behavior was not appropriate in the workplace. She's an attractive lady. Dr. Knight found her behavior and dress to be inappropriate."

For Nelson, a 32-year-old married mother of two, the news of her firing and the rationale behind it came as a shock.

"I was very surprised after working so many years side by side I didn't have any idea that that would have crossed his mind," she said.

The two never had a sexual relationship or sought one, according to court documents, however in the final year and a half of Nelson's employment, Knight began to make comments about her clothing being too tight or distracting.

"Dr. Knight acknowledges he once told Nelson that if she saw his pants bulging, she would know her clothing was too revealing," the justices wrote.

Six months before Nelson was fired, she and her boss began exchanging text messages about work and personal matters, such as updates about each of their children's activities, the justices wrote.

The messages were mostly mundane, but Nelson recalled one text she received from her boss asking "how often she experienced an orgasm."

Nelson did not respond to the text and never indicated that she was uncomfortable with Knight's question, according to court documents.

Soon after, Knight's wife, Jeanne, who also works at the practice, found out about the text messaging and ordered her husband to fire Nelson.

The couple consulted with a senior pastor at their church and he agreed that Nelson should be terminated in order to protect their marriage, Cochrane said.

On Jan. 4, 2010, Nelson was summoned to a meeting with Knight while a pastor was present. Knight then read from a prepared statement telling Nelson she was fired.

"Dr. Knight felt like for the best interest of his marriage and the best interest of hers to end their employment relationship," Cochrane said.

Knight acknowledged in court documents that Nelson was good at her job and she, in turn, said she was generally treated with respect.

"I'm devastated. I really am," Nelson said.

When Nelson's husband tried to reason with Knight, the dentist told him he "feared he would have an affair with her down the road if he did not fire her."

Paige Fiedler, Nelson's attorney, said in a statement to ABC News affiliate KCRG that she was "appalled" by the ruling.

"We are appalled by the Court's ruling and its failure to understand the nature of gender bias.," she wrote.

"Although people act for a variety of reasons, it is very common for women to be targeted for discrimination because of their sexual attractiveness or supposed lack of sexual attractiveness. That is discrimination based on sex," Fiedler wrote. "Nearly every woman in Iowa understands this because we have experienced it for ourselves."

Gene variants affect pain susceptibility in children

Dec. 24, 2012 ? At least two common gene variants are linked to "clinically meaningful" differences in pain scores in children after major surgery, reports a study in the January issue of Anesthesia & Analgesia, official journal of the International Anesthesia Research Society (IARS).

"[O]ur study is highly suggestive of a genetic component in pain response among children," concludes the study by Dr Chantal Mamie and colleagues of Geneva University Hospitals, Geneva, Switzerland. But an accompanying editorial question the relevance of this and previous studies of pain-related genes for management of pain in individual patients.

Gene Variants Influence Pain Scores after Surgery ?

The study was designed to explore whether several "candidate" gene variants affected pain scores in a group of 168 children undergoing major surgery -- either abdominal or bone and joint operations. The children and their parents were tested for variant forms ("polymorphisms") of six different genes previously reported as having a possible impact on pain.

The genetic results were compared with the children's pain scores, as routinely monitored during the 24-hour recovery period after surgery. During that time, the children had access to patient- (or parent- or nurse-controlled) analgesia with strong opioid (morphine-related) pain relievers.

Variants of two genes were related to "clinically meaningful" increases in pain scores -- at least four "peak" scores higher than six (on a ten-point scale) during the 24 hours after surgery. After adjustment for other factors, the risk of elevated pain scores was 4.5 times higher for children with a specific variant of the gene ABCB1, which affects the transport of opioid drugs to the central nervous system.

Risk of elevated pain scores was 3.5 times higher for children with a certain variant of the gene OPRM, a key target receptor for opioid binding. The associations with ABCB1 and OPRM variants remained significant after adjustment for patterns of gene inheritance from parents. Variants of two additional genes affecting pain perception -- NTRK and COMT -- were linked to more subtle, "subclinical" effects on pain scores.

? But Have No Effect on Use of Pain Medications

Surprisingly -- even though the gene variants affected pain scores -- they were unrelated to the total dosage of opioid medications used. The dosage of patient-controlled analgesia provides an important objective measure of pain and pain control after surgery.

"The present results are plausible given the known functionality of the candidate genes, and are consistent with the findings in adults," Dr Mamie and colleagues write. Although there has been a wealth of research on the genetic basis of pain in adults, the researchers add, "This first but small cohort study provides clues to further explore the genetic foundations of pediatric pain."

In an accompanying editorial, Drs Debra Schwinn and Ruth Landau of University of Washington, Seattle, put the findings in perspective. A decade ago, researchers thought that the discovery of genes affecting pain perception and opioid responses would soon play an important role in "individualizing" pain control after surgery. Subsequent studies have shown that the situation is more complex, and that the inheritance of pain susceptibility and opioid responsiveness is "probably less straightforward and predictable than previously foreseen."

Especially with the lack of effect on pain medication dosage, Drs Schwinn and Landau suggest that the presence of gene variants (genotype) may be less important than the way those genes are expressed in the individual (phenotype). Because of the complexity of the associations, they conclude, "[T]ailoring opioid analgesia based on selective genotyping is unlikely to occur anytime soon."
